چکیده

We created a rabbit model of retinoblastoma and confirmed the tumor clinically and histopathologically. Seventeen New Zealand rabbits were immunosuppressed with cyclosporin A at doses of 10-15 mg/kg. At day 3, the animals received a 30 μl subretinal injection of 1 x 10⁶ cultured WERI retinoblastoma cells. Digital fundus images were captured before euthanasia, and the eyes were submitted for histopathology. Retinoblastoma cells grew in all the inoculated eyes and established a tumor under the retina and/or in the vitreous. New blood vessels in the tumor were observed starting at week 5. Cuffs of viable tumor cells surrounded the blood vessels with regions of necrosis present at 70-80 μm from nutrient vessels. Occasional tumor seeds in the vitreous histologically exhibited central necrosis. This rabbit model demonstrated similar fundus appearance and pathologic features to human retinoblastoma and may be used as a model to test various routes of drug delivery for retinoblastoma.

Cytotoxicity effect of gold nanoparticles against a rabbit model of retinoblastoma

Introduction: Retinoblastoma is a malignant tumor in children. The goal of retinoblastoma treatment is to maintain vision and minimize side effects. In this study, the toxicity of the gold nanoparticle safety in vitro was investigated on an eye tumor of retinoblastoma under in vivo conditions. Materials and Methods: For the evaluation of the toxicity of gold nanoparticles using MTT test after 4...

Transducible peptide therapy for uveal melanoma and retinoblastoma.

OBJECTIVE To determine whether transducible peptides that inhibit the oncoproteins HDM2 and Bcl-2 may selectively kill uveal melanoma and retinoblastoma cells. METHODS Peptides were tested by viability assay, flow cytometry, TUNEL (terminal deoxynucleotidyl transferase-mediated fluorescein-dUTP nick-end labeling) assay, Western blot analysis, and reverse transcription-polymerase chain reactio...

Molecular Investigation of the Human Adenovirus in Retinoblastoma Patients

Background and Aims: Retinoblastoma tumors are the most common intraocular malignancy in childhood, leading to death after two years. The Human Adenovirus (HAdV) infection could be critical in the retinoblastoma pathogenesis due to the virus and retinoblastoma 1 interactions. The objective of the current study was to investigate the possible presence of the HAdV genome in the retinoblastoma pat...

A Review of Outstanding Retinoblastoma Researches on RB1 Genes: Focus on Treatment by Non-ionizing and Ionizing Radiation

Introduction: Retinoblastoma (RB) is a malignant retinal tumor, which affects infants and children. This cancer happens through a mutation in the retinoblastoma tumor suppressor genes (RB1). Inactivation of RB1 has been observed in more than 97% of all retinoblastoma patients with mutations. In recent years, RB treatment has developed significantly. RBchr('39')s treatment methods can be one or ...

Quantification of Mean Vessel Density in Retinoblastoma and Its Correlation with Local Tumor Invasion and Patients Survival

Background and Objectives: Retinoblastoma is the most common intraocular pediatric malignancy. Angiogenic factor expression such as VEGF (vascular endothelial growth factor) in retinoblastoma can be confirmatory angiogenic potential of this tumor. This study was performed to determine the role of angiogenesis in local invasion of retinoblastoma and its correlation with patients’ sur...
